Peeters's musical career and output have left a notable legacy in the world of organ and choral music. He was renowned for his deep knowledge of the late-romantic organ tradition, particularly inspired by César Franck, and for his integration of neoclassicism and polytonality. His works demonstrate a strong affinity for Gregorian chant and are characterized by their meditative and imaginative qualities.

Peeters composed nearly 500 works for organ, with many still frequently performed and widely respected for their craftsmanship and spiritual depth. Important works include "Passacaglia and Fugue" (1938), "Sinfonia" (1940), "Organ Concerto" (1944), "Lied Symphony" (1948), "30 Short Chorale Preludes" (1959), "213 Hymn Preludes for the Liturgical Year" (1959–1967), and "6 Lyrical Pieces" (1966). He also published esteemed collections and pedagogical works such as "Anthologia pro organo" (4 volumes, 1949–1959) and "Ars Organi" (3 volumes, 1952–1954).

Peeters held major teaching appointments at the Lemmens Institute, Royal Conservatory of Ghent, Tilburg Conservatory in the Netherlands, and Royal Flemish Conservatory of Music in Antwerp, also serving as director at the Antwerp conservatory from 1952 to 1968. He was named Baron by King Baudouin in 1971, a testament to his significant contributions to music.
